The native Victorian moved to Queensland with his family at 15-years-old and was drafted to the Lions just as their glory era was starting to peter out. He debut against the Demons in Round 10, 2004 and played eleven games, including a final, in his debut season before adding another 18 in 2005.

Early in 2006 Macdonald's career was disrupted when he suffered an ACL injury that kept him out until midway through 2007. He bounced back with 18 games in 2008 before playing every home and away match in 2009. The running defender was then dropped for the two Lions finals matches and parted ways with the club at the end of the season. He had requested a swap to a Melbourne club, but this had not eventuated during trade week.

Macdonald immediately began training with the Demons, and signed a two year deal to join the club as the first pick in that year's pre-season draft. He featured in the Demon side for the first six games of the season before being dropped, but bounced back late in the year to play a starring role across the backline in seventeen total games.

In and out of the side during 2011 he played just 10 matches, his best stay in the team being a six week stretch between Round 10 and Round 14.

After having three stints out of the side in the first half of 2012 Macdonald forged himself a permanent position in the backline and played every match of the season from Round 13 onwards. He received a contract extension at the end of the year. Macdonald had an injury hit 2013 campaign, battling hamstring, groin, and toe injuries before retiring in August after one match.

At the end of his career Macdonald launched business GetSwift with teammate James Strauss, before the relationship soured and Strauss sued him for $15 million.
