Melbourne used the second overall pick in the 2013 Rookie Draft on a lifelong Demons fan who had impressed with an outstanding TAC Cup playing in the Dandenong midfield. Before his TAC Cup career began, Harmes played eight games in the Devon Meadows senior team as a 16-year-old.

Debuted midway through 2015, playing eight of the last nine games of the season. In 2016 he consolidated his position in the side, playing 19 games and receiving a Rising Star nomination. Played a further 17 games the next year for 14 goals, used both in defence and attack.

Signed a two year contract extension in mid-2018, and had a career-best season. He played every game for the year, and became a vital cog in his side's midfield during the season including a number of high profile tagging jobs. He was prominent again in 2019, used more as a standard midfielder than a tagger, including 35 touches against Collingwood on Queen's Birthday.

Further extended his contract at the end of 2019, adding another four years after finishing fourth in the best and fairest. His 2020 was less successful, a move into defence failed to pay off and for the first time since becoming a full-time midfielder he found himself out of favour, and often out of the side.

Harmes broke his wrist in the opening game of 2021, missing six weeks. He remained in the side for the rest of the season, playing in the side's premiership victory. He had a less successful 2022, after a strong first half of the year he was dropped for two weeks, returning to perform a crucial tagging job in the Round 15 win over Brisbane Lions. He fell out of favour again late in the season, relegated to substitute twice before returning to the side in time to play in Melbourne's two losing finals. Courted by Essendon, he opted to stay with the Demons but had another frustrating year in 2023, appearing just nine times, three as a substitute.

At the end of the season Harmes was traded to the Western Bulldogs for their 2024 National Draft third round selection. He debuted against Melbourne, and was suspended for a week after headbutting premiership teammate Steven May.

Harmes represented Melbourne in the 2022 Grand Final Sprint.
