Acquired with the pick that the Demons received for trading Travis Johnstone to Brisbane, Grimes came to the club as an under-18 All-Australian.

Soon after, stress fractures in his back forced Grimes onto the long term injury list. After rehabilitation that included altering his running style, Grimes debuted in the last round of 2008.

There were fears that another back injury in the 2009 pre-season would keep him out all year but Grimes returned in Round 7, played 11 games and was nominated for the Rising Star award.

Grimes was a regular in 2010, playing the first 14 games before a hamstring injury in Round 14 ended his season. At the end of that year he signed a contract extension, despite having a year to run on his deal, to keep him at the club until the end of 2013.

Despite not having yet played a full season Grimes was elected to the leadership group for 2011. A broken navicular bone in his foot during the opening minutes of Round 6 left him out for the rest of the year.

Grimes was named co-captain for 2012 alongside Jack Trengove but suffered another setback when he hurt his shoulder in a training session. He returned for the start of the year and enjoyed his first ever injury free season, missing just one game through suspension, and was one of his side's best players in the second half of the season.

After his first injury free season Grimes started 2013 as one of his side's best in two diabolical performances. He then signed a contract extension to keep him at the club until the end of 2016, but suffered a broken collarbone in a tackle by former teammate Brock McLean during the Round 6 loss to Carlton and missed two months.

He played 18 games in 2014 despite three spells out of the side with injury, ending with a damaged hamstring in Round 21. After a further 12 games in 2015, Grimes struggled to break into an improving side the next year. After being named emergency several times he played two consecutive senior games in the middle of the game before being dropped again. He was delisted at the end of the year and returned to Hurstbridge.

Grimes' brother Dylan debuted for Richmond in the last game of the 2010 season, two years to the round after Jack made his debut against Richmond in round 22, 2008.
